HEPATOPROTECTIVE ACTIVITY OF ETHANOLIC EXTRACT FRACTIONS OF MOMORDICA CHARANTIA FRUIT, AGAINST ALCOHOL INDUCED HEPATOTOXIC RATS.

Abstract

The whole plant of Momordica charantia (M. C) Linn. (Cucurbitaceae) is used in variety of diseases in long-established Indian system of medicine including pancreatic and hepatic ailments. Cucurbitane Triterpenoids, the active constituents isolated from the plant Momordica Charantia fruit, was evaluated as a hepatoprotective agent against ethanol-induced hepatic injury in rats. LD studies for Momordica charantia fruit were conducted up to a dose 50 level of 2000 mg/kg dose as per OECD guidelines Number 425. Animals were administered orally with alcohol (3.75 g/kg for 10 days) after 1 hour of administration of three fractions (25, 50, and 100 mg/kg body weight) of Momordica charantia ethanolic extract for ten consecutive days except rats in normal and toxicant groups. The protective effect was evaluated by biochemical parameters including Aspartate transaminase (AST), Alanine transferase (ALT), Alkaline phosphatase (ALP), reduced Glutathione (GSH), Malondialdehyde (MDA) and Superoxide dismutase (SOD). The levels of these enzymes were found to be reduced in the cells with alcohol intoxication. Treatment with tree fractions of Momordica charantia fruit lead to significant amelioration of toxin-induced changes including Aspartate transaminase (AST), Alanine transferase (ALT), reduced Glutathione (GSH), Malondialdehyde (MDA) and Superoxide dismutase (SOD). Cucurbitane Triterpenoids treatment (Per orally for 10 days) restored the distorted parameters in a dose-dependent manner (36-100%).
