Combined inhibitory effect of sulforaphane and 5-flourouracil on cell proliferation in human ACC-M cells.

- Abstract
PURPOSE: To evaluate the growth-inhibitory effect of sulforaphane (SFN) and 5-fluorouracil (5-Fu) against the proliferation of human high metastatic cell line of salivary gland adenoid cystic carcinoma (ACC-M). The expression of nuclear factor kappa B (NF-κB) inducing resistance to anticancer chemotherapeutic agent was also detected. METHODS: The combination effect of SFN and 5-Fu was quantitatively determined using the method of median effect principle and the combination index. The NF-κB p65 expression after treatment with the SFN-5-Fu combination was also evaluated by Western blotting analysis. Statistical comparison was performed by Student's t test and analysis of variance (ANOVA), using SPSS 10.0 Software Package. RESULTS: The IC50s for SFN and 5-Fu in ACC-M cells were 38.58±4.97 μmol/L and 120.90±12.96 μmol/L, respectively. Treatment of ACCs cells with SFN and 5-Fu in combination, led to synergistic inhibition on cell growth and a decreased expression of NF-κB p65 protein. When the two drugs were combined, the level of NF-κB p65 protein expression was decreased almost 5-fold (P<0.05). CONCLUSION: The results demonstrate synergism between SFN and 5-Fu against the proliferation of ACC-M cells, which is associated with the decreased expression of NF-κB p65 protein.Supported by Postdoctorate Foundation of Heilongjiang Province(Grant No.LRD06-185).
