Isolation, Identification and Quantitative Determination of Ellagic Acid and Brevifolincarboxylic Acid in Duchesnea indica by RP-HPLC.

- Abstract
Duchesnea indica is a traditional Chinese medicine commonly used for treatment of cancers in combination with other herbs. Two phenolics, ellagic acid (EA) and brevifolincarboxylic acid (BA), were isolated from the phenolicfraction of the herb. In addition, a new RP-HPLC method for simultaneous determination of EA and BA was developed and validated. The separation was performed on a C18 column, gradiently eluted with 25 mM potassium dihydrophosphate buffer (pH 2.4) and acetonitrile at 1.0 mL/min and monitored at 360 nm. The plant materials were extracted with ethanol-water (50:50). The two components EA and BA showed good linearity (r² >0. 999) within the studied concentration ranges, with LOD of 12 and 20 ng/mL, respectively. The precision (RSD) was less than 1.50% for both two compounds and the extraction recoveries were 99.65% and 102.95% , for EA and BA, respectively. The developed method was applied to assay the two components in the herbs of Duchesnea indica.
