Inhibitory Effect of Polyphenol Extract from Pomegranate Peel on Aldose Reductase Activity.

Abstract

In this paper three polyphenol extracts from pomegranate peel were obtained using acetone, methanol and water as a solvent, respectively, and their inhibition on aldose reductase (AR) from bovine lens were determined on a ultraviolet spectrometry. All the extracts showed a dose-dependent inhibitory effect on AR activity. Of which, acetone extract demonstrated the strongest inhibition followed by methanol and water extracts, respectively, their 50% inhibition concentrations (µg/mL) on the enzyme being 34.77,44. 18 and 62.07. Compared with tea polyphenols, all the extracts showed a lower inhibitory capacity on AR. Acetone and methanol extracts exhibited an uncompetitive inhibitory effect on AR, and their inhibition constants were 49 and 71 µg/mL, respectively, at 0. 033 mg/mL concentration.
