Effects of Conjugated Linoleic Acid on the Proliferation and Differentiation of Preadipocyte 3T3-L1.

Abstract

3T3-L1 preadipocytes proliferation was detected by MTT method. Accumulation of lipid in the cytoplasm of differentiated adipocytes was observed by oil red O staining. The expression of peroxisome proliferators activated receptor γ2 (PPARγ2) mRNA was detected by RT-PCR. The t10, c12-CLA and two CLA mixture can significantly inhibit 3T3-L1 cell proliferation(P <0.01). Oil red staining showed t10, c12-CLA has strong inhibiting effect on 3T3-L1 cell line differentiation( P < 0.05). RT-PCR showed that 100 µmol/L t10,c12-CLA and 50 µmol./L t10,c12-CLA significantly reduced the expression of PPARγ2 mRNA as 52.1% and 83.0% in comparison with control group respectively.
