Structural Investigation of Oligosaccharides in Partial Acid Hydrolyzed Products of Fucoidan Isolated from Hizikia fusiforme.

Abstract

DSF32, a fraction of fucoidan isolated from Hizikia fusiforme, was partially hydrolyzed by 0.5 mol/L trifluoroacetic acid. The hydrolyzed product was dialyzed using a dialyzer with a Mw cutoff of 3500 Da. The permeance of dialysis was separated into two parts of neutral sugars (05N) and acid sugars (05A) by D201 column. The structures of 05N and 05A were studied by HPLC/MS and methylation analysis. The results showed that 05A may contain the following oligosaccharides:→4) GlcA (1→2) Hex (1→,→2) Hex (1→4) GlcA (1→2) Hex (1 → and →4) GlcA (1→2) Hex (1→3) Fuc(1→.05N-3, a component of 05N, has a non-reducing end of Hex (1→ and a reducing end of 2→) Hex, and the types of glycosidic bonds in oligosaccharides are mainly 1,6 plus a small amount of 1,4 and 1,2,6.
