Optimization of ultrasonic assisted extraction of pleurotus eryngii polysaccharide by response surface methodology and its antifatigue activity.

Abstract

Pleurotus eryngii was extracted by ultrasonic--assisted extraction and its anti--fatigue effect was studied. The extraction rate of pleurotus eryngii polysaccharide was used as the index, extraction process was optimized by response surface method on the basis of single factor experiment. The optimum conditions were as follows : ultrasonic power 420 W, ultrasonic temperature 50 C and ultrasonic time 50 min. Under the above conditions, the extraction rate of pleurotus eryngii polysaccharide was 14.71%. The anti--fatigue effect of pleurotus eryngii polysaccharide was measured by observing the time of mice climbing and weight--bearing swimming, serum biochemical indexes, and liver glycogen and myoglycogen. The results showed that pleurotus eryngii polysaccharide could significantly increase the climbing and swimming time of mice, SOD activity, reduce La content, and increase the content of liver glycogen and muscle glycogen, which indicating that pleurotus eryngii polysaccharide has anti-- fatigue effect.
