Optimization of purification process of flavonoids of Gardenia flower and its anti-oxidant and anti-fatigue effects.

Abstract

The flavonoids extract of Gardenia flower was purified by macroporous resin, the optimum type of macroporous resin was determined by comparison of static adsorption-desorption performance in five type macroporous resin. The optimum purification condition was explored by single factor experiment. The effects on antioxidant and anti-fatigue of different products were investigated by radical scavenging in vitro and animal tests. The results showed that the optimum purification conditions were as follows: the loading concentration was 6.0 mg/mL and loading solution pH was 5.0, the loading flow rate was 1.0 mL/min, the volume of elution solution ethanol was 140mL and elution flow rate 2.0 mL/min. The purity of product increased to (76.7±1.2) % with the optimum process. The IC50 of purified Gardenia flower flavonoids on DPPH free radical and hydroxyl radical was lower than the extract. Purified Gardenia flower flavonoids could obviously improve the activities of LDH, GSH-Px and SOD, reduce the concentration level of lactic acid and malondialdehyde, and prolonged exercise time of mice, had better anti-fatigue effect. Therefore, it can be used as raw material for sport food development.
