Design and In Vivo Characterization of Novel Pulsatile Drug Delivery System of Biguanide Anti-Diabetic.

Abstract

The aim of the current work was to design and develop pulsatile pellets of hydroxypropyl cellulose and eudragit in an admixture for a gastro-retentive drug delivery system. Pellets containing metformin hydrochloride (500 mg) were prepared by the pelletization technique using an extruder-spheronizer. Metformin hydrochloride spherical pellets were prepared, It was characterized by complete release of the drug in pulses after well-defined lag time of 6 h (± 0.20) (period of no drug release) for the treatment of diabetes mellitus. The novel drug delivery system consisted of HPC Type H as lag time modifier, and Eudragit L-100 and Eudragit S-100 in different concentration as pH dependent release modifier in gastrointestinal tract. The pellets of the selected batch were having spherical and smooth surface with maximum % yield of 78.98 %.The pellets were subjected to in vivo studies using HPLC and LC-MS for the optimized formulation. The AUC0 t, Tmax, Cmax, Cl, Vd was found to be 345.5, 53.6, 1471.3, 0.187, 89.9 respectively. The selected formulation batch-5 consisted of HPC Type in concentration (18 %); Eudragit L-100 (29 %) and Eudragit S-100 (19 %) w/w respectively.
