DEVELOPMENT AND VALIDATION OF ANALYTICAL HPLC METHOD FOR SIMULTANEOUS ESTIMATION OF AMOXICILLIN AND PROBENCIDE IN TABLET.

Abstract

A simple, accurate, rapid, economical RP-HPLC method have been developed and validated for estimation of Amoxicillin and Probencide in combined tablet dosage form and can be used in routine analysis. In RP-HPLC, method was carried out by isocratic technique on a reversedphase: by C18 column (250×4.6 mm, 5 μm Particle Size) column kept at ambient temperature and with mobile phase containing a mixture of buffer (pH4.0)-methanol (40:60% v/v) at a flow rate of 1.0 ml/min. The average retention times for Amoxicillin and Probencide were 3.447min and 6.193min respectively. The calibration curves were linear in the concentration range of 5-15μg/ml for Amoxicillin and of 5-15μg/ml for Probencide. Correlation coefficients were found to be 0.9976 and 0.9985 for Amoxicillinand Probencide respectively. Mean assay was found to be 99.95% and 102.73% for Amoxicillin and Probencide respectively. All the three methods were compared statistically using ANOVA and it was found that they are not significantly differ indicating their suitability in routine analysis.
