Abstract

Objective To explore the effect of kidney-tonifying, spleen-strengthening, and blood stasis removing recipe on the expression of ERRa and PGC-1α mRNA in the bone marrow of the rat femurs in an ovariectomized rat model. Methods Forty 7-month-old female SD rats were randomly divided into sham-operated group, ovariectomized (OVX) group, herb group (kidney-tonifying, spleen-strengthening, and blood stasis removing recipe), and alendronate (ALEN) group, with 10 rats per group. After a 12-week treatment, the bone mineral density (BMD) of the left femur was measured using small animal special dual energy X-ray absorptiometry. Bone morphology of the proximal tibia was observed using HE staining. The expression of ERRa and PGC-1α mRNA of bone marrow was detected using quantitative PCR. Results Compared with that in sham-operated group, BMD in OVX group was obviously decreased (P 0.05). Conclusion The kidncy-tonifying, spleen-strengthening, and blood stasis removing recipe enhances rat BMD by increasing the expression of PGC-1α mRNA and decreasing the expression of ERRα mRNA.
