- Title
Biologic Structure and Function: Perspectives on Morphologic Approaches to the Study of the Granular Layer Keratinocyte.

Holbrook, Karen A.
- Abstract
The development of morphologic approaches for skin research has involved the development of instrumentation methods to prepare tissue for morphologic examination and techniques to preserve and process specimens in as natural a state as possible, free of distortion and artifact. Development of methodology for tissue preparation is as important as the development of microscopes with high resolving power, the use of sophisticated instruments to examine skin specimens is irrelevant lithe tissue is poorly preserved and thus provides only limited information. This can be appreciated by noting the lack of clarity and detail in the electron micrographs of the investigators in the 1950s and comparing them with the relatively high resolution images of cell structure obtained by investigators in even the late 1960s.
