- Abstract
Elaeocarpus variabilis a well-known medicinal plant of Elaeocarpaceae family is used by many tribal groups to treat inflammation in India. In the study, different extracts such as ethyl acetate, ethanol and Aqueous of E. variabilis leaves and stem are evaluated for its secondary metabolites, in-vitro antioxidant, antibacterial and anti-inflammatory activity and anticancer. Among various extracts, ethyl acetate extract of stem showed highest content of total phenolics (154.09 mg GAE/ g extract), tannin content (62.41 mg GAE/g extract) and flavonoid content (173.84 mg RE/g extract). In the present study, in vitro antioxidant assays were carried out ethanol extract of leaf showed highest DPPH radical scavenging assay (IC50 values was (100.4 µg/mL), ethyl acetate extract of stem showed highest results in phosphomolybedenum assay (293.13 mg AAE/g), ferric reducing assay (391.66 mM Fe (II) E/mg extract), ABTs assay (182014 µg TE/g) and nitric oxide assay (54.26%). Among the various in-vitro antioxidant assays carried out in different extracts, the ethyl acetate and ethanol extract exhibited higher antioxidant activity in all the assays. Moreover, ethanol extract of leaf exhibited good antibacterial activity against Klebsiella pneumonia (14 mm), Pseudomonas aeruginosa (13 mm), Streptococcus pneumoniae (14 mm) and Staphylococcus aureus (19 mm), at 20 mg/ml concentration. The maximum membrane stabilization activities were exhibited by the ethyl acetate stem extract of E. variabilis (95.16%) followed by ethanol extract of leaf of hypotonic or heat-induced activity (117.28 µg/mL). In invitro cytotoxicity assay, E. variabilis stem ethyl acetate extract decreases the cell proliferation activity. The IC50 cell inhibition value for the E. variabilis stem ethyl acetate extract was 63.68 ìg/ml. E. variabilis stem extracts have a tremendous amount of antioxidant potential, making them a good source of natural antioxidant supplements for food to protect against oxidative stressrelated diseases, including anti-cancer.
