- Abstract
We proposed CCA-1.1, one of the curcumin analog derived from PGV-1 to be explored for anticancer activities toward luminal MCF-7 breast cancer cells, covering on the cytotoxic activity in correlation with cell cycle inhibition, apoptosis induction, ROS generation, and senescence evidence. The cell viability was conducted by MTT assay; cell cycle profile, apoptosis, and ROS level were measured based on the fluorochrome through flowcytometry-based assay; and senescence phenomenon was evaluated using SA-ß-gal assay. CCA-1.1 performed cytotoxic activity similarly with PGV-1 against MCF-7 cancer cells. Interestingly, CCA-1.1 induced rapid cell death and inhibited cell cycle progression particularly at G2/M, and tended to induce apoptosis. Moreover, CCA-1.1 and PGV-1 also increased intracellular ROS level, which attributed to cell senescence and led to cellular death. Taken together, our preliminary study reinforces the potency of CCA-1.1 based on its anticancer activities against luminal breast cancer cells and prospective to be pharmaceutically developed as a chemotherapeutic agent.
