- Abstract
Coreopsis tinctoria Nutt. extracts were prepared using water and ethanol at concentrations of 30%, 50%, 70%, and 90%, followed by the combination of the extracts to evaluate the in vitro antibacterial activity. Subsequently, the extract was administered via gavage to mice subjected to chronic unpredictable mild stress (CUMS) to investigate its effects on four types of intestinal bacteria. The results indicated that the total flavonoid content of the combined extract was 30.00%, with a malicin content of 0.83%. In vitro analysis demonstrated the extracts exhibited significant antibacterial activity against Escherichia coli, Salmonella, and Staphylococcus aureus, with minimum inhibitory concentrations of 0.625, 0.625, and 0.3125 mg/mL, respectively. Administration of the extract to CUMS depression model mice not only reversed the depressive behaviors induced by stress but also inhibited the growth of Enterococcus and E. coli, while promoting the proliferation of Lactobacillus and Bifidobacterium, thereby enhancing gut colonization (B/E ratio) . In conclusion, Coreopsis tinctoria Nutt. extract demonstrated effective in vitro antibacterial properties and positively influenced depressive behaviors and gut microbiota in CUMS depression model mice.
