The UraA and RutG transporters of <italic>Escherichia coli</italic> are members of the ubiquitous Nucleobase‐Cation Symporter‐2 (NCS2) family. Structure and function comparisons identify protein sequence differences that determine selectivity. Replacement of Phe73 with Ala in the UraA uracil‐specific permease allows binding of thymine with an affinity that is comparable to the broad‐specificity uracil/thymine permease RutG.
