The article focuses on the synthesis of keratan sulfate, a chain of glycan, with disaccharide unit Galßl-4GlcNAcß. Topics discussed include conjugation of disaccharids with biotin through hydrophilic linkers, coupling of glycosyl acceptor with trimethylsilyl triflate (TMSOTf) and analysis of synthesis of keratan sulfate through silica gel chromatography, thin layer chromatography and nuclear magnetic resonance. An overview of the chemical compounds used in the process is also given.
