- Abstract
A strain SLLSM2 with high yield of sucrose phosphorylase was isolated and screened from the soil of Laibin sugarcane field, and the strain was identified as Bacillus velezensis by 16S rDNA. The enzyme production conditions were optimized by single factor test and response surface test using the strain SLLSM2 as starting strain, and then the α-arbutin synthesis conditions catalyzed by the enzyme were optimized. The results showed that the enzyme production reached the highest under the conditions of sucrose content 7.5%, peptone content 9.0%, inoculum 10%, pH 6.0 and fermentation time 96 h, and the sucrose phosphorylase activity increased from 23.12 U/ml to 120.07 U/ml, which was 5.19 times higher than that before optimization. The optimal conditions for the synthesis of α-arbutin catalyzed by the enzyme were as follows: enzyme addition 150 U, hydroquinone concentration 4%, reaction temperature 45 ℃, pH6.0. Under the optimal conditions, the molar conversion of α-arbutin synthesis reached 44.09%.
