- Abstract
In the present investigation the practical application possibilities of the new GenePath Strain Typing System® for molecular biological DNA finger printing were demonstrated by the analysis of different bacterial strains, most of them isolated during the course of nosocomial infections. By commercially available reagent-sets, including restriction enzymes, proteinases, buffer and culture media as well as agarose, control plugs and control cultures, a standardized pulsed-field gel electrophoresis (PFGE) for the clonal differentiation of bacteria for diagnostic and epidemiological purposes is possible. The study of intra- and inter-assay precision showed the satisfying reproducibility pf the results. Within three days the clonal relationship among various bacterial isolates can be proved by this form of macrorestrictipii analysis. System handling, application of the reagents as well as documentation and interpretation of the restriction fragment patterns on the ethidium bromide stained gels are pretty simple and easy to learn. Pictures of stained restriction fragment patterns show the clonal identity of isolated bacteria and give by this way evidence for nosocomial infections.
