- Abstract
The article discusses the result of the study regarding the contribution of a single free cysteine residue and disulfide bond to the thermostability of Aspergillus saitoi 1,2-α-mannosidase, in Japan. A. saitoi 1,2-α-mannosidase contains three conserved cysteine residues, which were not essential for the activity analyzed by site-directed mutagenesis and kinetics. The destabilization of the enzyme was due to the substitution at each cysteine residue. Thus, the hydrophilic and large side chain causes the destabilization.
