RESVERATROL TREATMENT UPREGULATES GLUT-4 TRANSLOCATION TO THE CAVEOLAR MEMBRANE DOMAINS THROUGH AKT / eNOS SIGNALING IN RAT MYOCARDIUM.

- Abstract
Homeostasis of blood glucose by insulin (I) involves rapid stimulation of glucose uptake by translocation of glucose transporter Glut-4 from intracellular pool to the caveolar membrane system. Impaired insulin resistance & glucose transport generally characterize diabetes. In the present study we examined resveratrol (R) a polyphenol from redwine mediated regulation of glucose levels, Glut-4 translocation & its interaction with caveolin-1 & 3 (Cav-1 & 3) in the streptozotocin (STZ) induced diabetic myocardium. The rats were grouped as Control (C), Diabetic (D) (STZ 65mg/Kg, i.p) & D + R (2.5mg/Kg b.w for 2 weeks orally) (DR). Decreased blood glucose level was observed in DR (370mg/dl) when compared to D (>500mg/dl) which was confirmed with H9c2 cardiomyoblasts. Isolated lipid raft fractions in D has shown decreased expression of Glut4, Cav3 (0.4, 0.6 fold) & increased Cav1 expression (1.4 fold) compared to C. R treatment has shown increased expression of Glut4, Cav3 (0.75, 1.1 fold) and decreased expression of Cav1 (0.7 fold) compared to D. Increased phosphorylation of eNOS and Akt was observed in the cytosolic fractions of DR compared to D. Significant association of Glut-4 & Cav-3 was observed in R compared to D. Thus, we conclude that increased activation of Akt, Glut-4 & Cav-3 association and disruption of caveolin-1 / eNOS interaction results in glucose uptake during R treatment in the diabetic myocardium.
