- Abstract
Stipe is the main edible part of Flammulina filiformis and other mushrooms. However, the mechanism of its growth and development is still unclear. The transcriptome and proteome of the elongating and maturated stipes of F. filiformis were analysed. Then, the differential expression genes (DEGs) and differential expression proteins (DEPs) were analyzed based on gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) database. Comparison of gene expression patterns in the maturation and elongation stipes revealed that DEGs and DEPs were respectively 721 and 61. Most of them were up-regulated. 72.14% DEGs were clustered in catalytic activity term based on GO database. Lots of DEGs and DEPs were clustered in cell part and binding terms. Most DEGs were clustered in carbohydrate metabolism and amino acid metabolism based on KEGG database. However, most DEPs were clustered in monobactam biosynthesis, streptomycin biosynthesis and selenocompound metabolism pathways. Protein processing in endoplasmic reticulum and MAPK signaling pathway were the differential pathway with both DEGs and DEPs. In this study, 40 candidate genes in stipe development process of F. filiformis were obtained. It is helpful to reveal the stipe development mechanism of edible mushrooms.
