- Abstract
Objetivo: analizar la función supresora de las células Treg en pacientes con EP. Antecedentes: la enfermedad de Parkinson (EP) es el segundo padecimiento neurodegenerativo más frecuente. La EP, causada por la muerte de neuronas dopaminérgicas de la sustancia nigra, se asocia con cambios en la respuesta inmune en el sistema nervioso central y en la periferia. Además del proceso inflamatorio, se ha planteado que una deficiencia en la respuesta inmune reguladora podría subyacer a la neurodegeneración, y los pacientes muestran niveles menores de células T reguladoras (Tregs). No obstante, los hallazgos sobre la funcionalidad de las Tregs en pacientes con EP son contradictorios. De confirmarse, la reducción de Tregs podría favorecer el proceso inflamatorio en los pacientes con EP, y el avance de la enfermedad. Métodos: se reclutaron 4 pacientes con EP y 8 sujetos control. En una muestra de sangre periférica se marcaron células efectoras (CD4+CD25-CD127+/-) y Tregs (CD4+CD25hiCD127-). Las células se purificaron utilizando un citómetro sorter. Para analizar la capacidad de suprimir la respuesta proliferativa, se cultivaron ambas poblaciones con distintas proporciones de Tregs:células efectoras (1:1, 1:2, 1:4,1:8,1:16, 1:32) y se indujo la proliferación celular utilizando anticuerpos anti CD3/CD28. La proliferación de la población efectora se midió mediante tinción con efluor 670, y su inhibición se evaluó por citometría de flujo. Núm. de registro del protocolo: 145/19 Resultados: se observó una menor función supresora de Tregs en pacientes con EP que en el grupo control desde la relación Tregs:efectoras 1:1 (p=0.014). La disminución en la función supresora crece conforme disminuye la relación Tregs: efectoras. Conclusiones: se redujo la capacidad de las Tregs para inhibir la proliferación de células efectoras en pacientes con EP. Este hallazgo podría relacionarse con un defecto en la supresión de la respuesta inflamatoria que caracteriza a la EP, contribuyendo al avance de la enfermedad.
