- Abstract
Süperoksit dismutazlar (SOD), süperoksidin oksijen ve hidrojen perokside dismutasyonunu katalizleyen bir enzin sınıfıdır [1]. Bu enzimler oksijene maruz kalan hemen hemen tüm hücrelerde antioksidan mekanizmada önemli rol oynarlar. İnsanlarda farklı genlerce kodlanan üç SOD izoformu eksprese edilmektedir. SOD1 (CuZn-SOD) ağırlıkla sitoplazmada bulunurken, SOD2 mitokondride ve SOD3 ise ekstraselüler de lokalizedir. SOD1, birçok memeli hücresinde toplam hücresel SOD aktivitesinin ~ 85%' ini oluşturmaktadır [2]. SOD genetic polimorfizmi, kanser ve diğer hastalıklara yatkınlıkla ilişkilendirilmiştir. Tek nükleotid polimorfizmi içeren (SNP) -251A/G (rs2070424) SOD1 gen, oksidatif stresle ilgili bu potansiyel olarak önemli genetic variantlar kapsamlı olarak çalışılmıştır [3,4]. Bu çalışmada, 494 (kadın: 278, % 56,3 ve erkek: 216, % 43,7) sağlıklı bireyi içeren Türk popülasyonunda, PCR-RFLP metodu kullanılarak SOD1 gene -251A/G polimorfizmin dağılımı araştırıldı. Tüm genotip ve allellerde Hardy-Weinberg dengesindedir (X2=0.107, p>0.05). Çalışma popülasyonunun yaş ortalaması 38,4±16,6 (erkek: 39,8±17,1; kadın: 37.3±16.1) bulundu. A ve G allel frekansları sırasıyla 0.93 and 0.07 saptandı. Gözlenen genotip frekansları AA, AG ve GG için sırasıyla 86.2, 13.4 ve 0.4 % bulundu.
