- Abstract
Objective: A rapid, inexpensive and high-throughput assays for drug susceptibility testing (DST) of Mycobacterium tuberculosis is urgently required. For this reason, a direct microplate-based colorimetric assay which excludes the use of pre-testing culture isolate was evaluated. Design: This study is cross sectional study. Materials and Methods: TEMA was performed on 41 acid fast bacilli positive sputum specimens by direct inoculation of the processed specimens into the microplate containing serial-diluted first-line anti-tuberculosis drugs. One-Way ANOVA and Kruskal-Wallis analysis was used to evaluate the turnaround time. Results: Results from direct TEMA were obtained for isoniazide and streptomycin, with overall specificity, sensitivity and accuracy of 95%, 100% and 95% respectively. However, results obtained with rifampicin and ethambuthol were less encouraging with an overall specificity, sensitivity and accuracy of 61%, 88% and 63% respectively. A shorter mean turnaround time of 14 days was observed for direct TEMA (p < 0.001). Conclusion: Thus direct TEMA is potentially rapid, reliable and inexpensive method for DST of M. tuberculosis in countries which are often burden with high prevalence rates of multidrug-resistant tuberculosis.
