- Abstract
The experiment was to explore the optimal conditions for the production of cellulase by Bacillus velezensis. The DNS method was used to determine the activities of cellulase and filter paper enzyme. The results showed that when the inoculation amount was 8%, the cellulase and filter paper enzyme activities were the highest. When the fermentation time was 72 h, the temperature was 45 ℃, the environment pH value was 6.6, the cellulase activity was the highest. When cellobiose was used as the carbon source nutrition, the activity of cellulase reached the maximum, the fermentation time was 48 h, the temperature was 45 ℃, and the pH value of the environment was 5.4. When the filter paper enzyme activity was the highest. When starch was used as the carbon source nutrition, the activity reached the maximum. The experiment indicates that the optimal conditions for cellulase production by Bacillus velezensis are 8% inoculum, fermentation time 72 h, temperature 45 ℃, initial environmental pH value of 6.6, and the most suitable carbon source is cellobiose. The optimal conditions for producing filter paper enzyme activity are 8% inoculum, fermentation time 48 h, temperature 45 ℃, initial environmental pH value of 5.4, and the most suitable carbon source is starch.
