Efeito da Fitocistatina CsinCPI-2 sob a progressão da doença periodontal e artrite experimental. Estudo pré-clínico em camundongos.

- Abstract
As cistatinas são inibidores naturais das peptidases da cisteína. As proteínas CsinCPI-2 de Citrus sinensis foram identificadas e produzidas recombinantemente pelo nosso grupo. Estudos prévios identificaram efeitos inibitórios sobre a atividade de Catepsina B, K, IL-1ß e TNF-α. Sendo assim, o objetivo deste estudo é avaliar in vivo o papel da fitocistatina CsinCPI-2 na progressão da doença periodontal (DP) e artrite reumatoide (AR) induzida por antígeno (AIA) em camundongos. Material e Métodos: Foram utilizados 52 camundongos C57BL-6 que foram divididos em 3 estudos. O 1º estudo avaliou o efeito da CsinCPI-2 sobre a progressão da DP induzida por ligadura. Os animais foram distribuídos em dois grupos experimentais (n=6): Controle (sem DP) e teste (com DP). CsinCPI-2 foi administrada diariamente após a colocação da ligadura. Os animais foram sacrificados após 15 dias para avaliação da perda óssea alveolar por micro-CT. No 2º estudo, 25 animais foram divididos em 5 grupos: 1-Naive; 2-AIA (artrite induzida por antígeno); 3-AIA + 0.8ug/g CsinCPI-2; 4-AIA + 1.6ug/g CsinCPI-2; e5-AIA+ 3.2ug/g CsinCPI-2. Os parâmetros clínicos de dor, edema, e migração de neutrófilos foram avaliados. No 3º estudo, 15 animais foram distribuídos em 3 grupos: 1-Naive; 2-AIA; e 3-AIA + 3.2ug/g CsinCPI-2. Análise histológica foi realizada para avaliação da inflamação, morte de condrócitos, e erosão óssea. Resultados: CsinCPI-2 reduziu a perda óssea alveolar na maxila e foi efetiva na redução da dor, do edema, e da migração celular de neutrófilos. Na análise histológica foi verificado uma diminuição da inflamação, na morte de condrócitos e na erosão óssea na articulação tibiofemoral. Conclusão: CsinCPI-2 desempenha um papel importante na diminuição da perda óssea alveolar em camundongos com DP induzida por ligadura, e na redução dos sinais clínicos e histológicos de artrite experimental.
