- Abstract
La mosca blanca Bemisia tabaci L. transmite begomovirus a cucurbitáceas en la Comarca Lagunera de Coahuila y Durango, México. Bemisia tabaci es una plaga que causa daños al alimentarse de la savia de las plantas y tiene la capacidad de transmitir virus, se caracteriza por ser un complejo de especies cripticas morfológicamente indistinguibles que pueden ser reconocidas por medio de técnicas moleculares. Durante 2015 y 2016, se analizaron moscas blancas para identificar los biotipos B, Q y nuevo mundo por PCR múltiple usando primers específicos basados en el gen mitocondrial citocromo oxidasa I. También se determinó la presencia de begomovirus en B. tabaci y en plantas en localidades productoras de Coahuila y Durango, mediante el uso de PCR con oligonucleotidos degenerados. Las secuencias obtenidas en el presente trabajo al ser comparadas con la base de datos del GenBank del NCBI, corroboraron la identificación del biotipo B con una similitud del 100% con secuencias tipo. En las muestras de 2015 se determinó que el begomovirus predominante en las moscas blancas fue el virus del mosaico dorado del chile (PepGMV) con 100% de similitud; mientras que en 2016 prevaleció el virus de la hoja arrugada de las cucurbitáceas (CLCrV) con un 97% de similitud. Bemisia tabaci is one of the most threatening pests of solanaceum and cucurbit crops in Coahuila and Durango, Mexico. It transmits viruses and adapts to different hosts and environments, creating a rapid dissemination and persistence. B. tabaci is characterized as a complex of cryptic species composed of morphologically indistinguishable biotypes that can be recognized by molecular techniques. During 2015 and 2016, B. tabaci specimens were analyzed to identify B, Q, and New World biotypes using multiplex-PCR and specific primers based on the mitochondrial cytochrome oxidase I gene. The presence of begomovirus in insects and plants collected in Coahuila and Durango fields was also determined. The sequences obtained were compared with the NCBI GenBank database, corroborating biotype B with a similarity of 100% with type sequences. In 2015, the begomovirus was determined with 100% to be the Pepper golden mosaic virus (PepGMV), and with 97% similarity to be the cucurbit leaf crumple virus (CLCrV) in 2016.
