EFEITOS DA HIPÓXIA-ISQUÊMICA PRÉ-NATAL E DA SEPARAÇÃO MATERNA NO COMPORTAMENTO TIPO-DEPRESSIVO DE RATOS.

- Abstract
Introdução: Eventos estressantes no período perinatal podem ser determinantes para a gênese de patologias, tendo em vista que esse período é fundamental para a maturação e o desenvolvimento dos sistemas biológicos. A presença de estressores no período perinatal tem sido associada ao aumento da susceptibilidade a desordens psiquiátricas, como o transtorno de depressão maior (TDM). Tendo em vista a importância do período neonatal para o indivíduo, existem na literatura modelos experimentais que buscam investigar os efeitos associados ao estresse e mimetizar o que ocorre em humanos, como exemplo da hipóxia isquêmica pré-natal (HI) e da separação materna (SM). Objetivo: O objetivo do presente estudo é avaliar os efeitos da hipóxia-isquêmica pré-natal (HI) e da separação materna (SM), isoladamente e em combinação, no comportamento tipodepressivo. Método: No 18° dia gestacional, fêmeas Wistar grávidas foram divididas aleatoriamente em grupo controle (CN), controle cirúrgico (SHAM) e hipóxia-isquêmica (HI). As ratas HI tiveram seus cornos uterinos expostos e suas artérias uterinas foram clampeadas durante 45 minutos. No grupo SHAM, as fêmeas foram submetidas ao mesmo procedimento, exceto pelo clampeamento das artérias uterinas. Após o nascimento a termo dos filhotes, as ninhadas dos três grupos foram subdividas em não separadas e separação materna. O protocolo de SM ocorreu do primeiro ao décimo quinto dia pós-natal por 180 minutos. Em P30 e P90, os animais dos seis grupos foram submetidos aos testes de preferência a sacarose e nado forçado para investigar o comportamento tipo-depressivo.(Protocolo CEUA 056/2018). Resultados: A hipóxia-isquêmica e a separação materna per se parecem gerar comportamento de anedonia nos animais adultos, padrão também observado nos animais submetidos à combinação dos estresses. De forma semelhante, no teste do nado forçado, a HI e a SM, de forma isolada e combinada, foram capazes de aumentar o tempo de imobilidade e reduzir a mobilidade nos animais, parâmetros esses relacionados ao desenvolvimento de comportamento tipo-depressivo. Conclusão: O conjunto de dados sugere que tanto a hipóxia-isquêmica, a separação materna precoce e a combinação desses dois modelos produzem alterações permanentes nos animais, que são consistentes na susceptibilidade a transtornos psiquiátricos, confirmando a hipótese de que eventos estressantes nas fases críticas do desenvolvimento podem ser determinantes para o surgimento de futuras patologias. O modelo empregado pode apontar caminhos para a investigação de possíveis intervenções farmacológicas.
