Kristall‐Engineering: ein Blick in die Zukunft.

- Abstract
Kristall‐Engineering befasst sich traditionell mit Molekülkristallen. Intermolekulare Wechselwirkungen im Zusammenhang mit der Kristallpackung werden aufgeklärt, und die so erhaltenen Kenntnisse werden für den Entwurf neuer Festkörper mit gewünschten physikalischen und chemischen Eigenschaften genutzt. Wir besprechen fünf Themenbereiche, die in den Umfang des Kristall‐Engineerings fallen und für die wir annehmen, dass sorgfältig geplante Forschungsarbeiten zu einem erheblichen Gewinn für die Wissenschaft mit Nutzen für die Gesellschaft führen könnten. Wir berühren Themen und Gebiete für künftige Forschungsförderung und Translationsbemühungen vor dem Hintergrund aktueller gesellschaftlicher Entwicklungen, bei denen zunehmend Anwendungen und nutzbare Produkte erwartet werden. Die fünf Themenbereiche sind: 1) pharmazeutische Feststoffe; 2) industrielle Festkörperreaktionen; 3) mechanische Eigenschaften mit praktischen Anwendungen; 4) Festkörper mit MOF‐ und COF‐Gerüsten; 5) neue Materialien für die Solarenergiegewinnung und fortgeschrittene Polymere. Gute Aussichten: In den letzten drei Jahren wurden von der USFDA zwei Kokristall‐Salz‐Wirkstoffe zugelassen, die mit Kristall‐Engineering entwickelt worden sind. Wenigstens drei weitere Wirkstoffkandidaten befinden sich im behördlichen Entscheidungsprozess. Für das Problem der schlechten Löslichkeit, das beinahe 90 % der Wirkstoffe in den BCS‐Klassen II und IV betrifft, bieten pharmazeutische Kokristalle einen Lösungsansatz.
