- Abstract
This article discusses the use of cryo-electron tomography (cryo-ET) and subtomogram averaging to explore biomolecules within their natural environment, a method known as visual proteomics. The study focuses on Chlamydomonas reinhardtii, a unicellular green alga, and aims to unravel high-resolution cellular structures. The article highlights the challenges faced by cryo-ET, such as low signal-to-noise ratio and increased motion during imaging, and introduces the Electron Event Representation (EER) image format as a solution to preserve data quality. The authors are currently testing the benefits of EER format and plan to compare it with conventional TIFF format. The study also includes the creation of an open access database for C. reinhardtii tomograms to facilitate further research and development of computational tools.
