Citotoxicidade e efeito estético de um scaffold contendo TiF4 aplicado como estratégia catalisadora no clareamento dental de consultório.

- Abstract
Introdução: Lesão pulpar e dor pós-clareamento dental convencional de consultório (CDCC) estão relacionados com a difusão trans-amelodentinária de altas doses de H2O2 residual liberadas dos géis clareadores. Assim, estratégias para catalisar o H2O2 têm sido propostas para prevenir estes efeitos adversos. Objetivos: Formular um scaffold (SC) contendo tetrafluoreto de titânio (TiF4) e avaliar sua influência sobre a eficácia estética (EE) e citotoxicidade (CT) causadas pelo CDCC. Material e método ou Conduta Clínica: Discos de esmalte/dentina foram adaptados em câmaras pulpares artificiais. Então, um gel clareador com 40%H2O2 foi aplicado por 20 min. (1 sessão) sobre o esmalte revestido ou não com um SC contendo ou não TiF4, estabelecendo os seguintes grupos: G1: sem tratamento (controle negativo); G2: 40%H2O2 (controle positivo); G3: SC+40%H2O2; G4: SC+TiF4+40%H2O2. A incorporação de TiF4 no SC foi avaliada por FTIR, sendo suas características analisadas por MEV e EDS. A EE (ΔE00 e ΔWI; n=4) e a quantidade de H2O2 difundido pelos discos (violeta-leucocristal/peroxidase; n=8) foram determinadas logo após os tratamentos. Os extratos (meio de cultura + componentes do gel clareador difundidos) foram aplicados sobre células MDPC23, as quais foram avaliadas quanto a viabilidade (Vi - Alamar Blue; n=8 e Live/Dead; n=4) e estresse oxidativo (EOx; n=8). Os dados foram submetidos ao teste ANOVA/Tukey, com nível de significância de 5%. Resultados: A incorporação de TiF4 no SC originou um biomaterial permeável, composto por fibras uniformes e regularmente distribuídas. Em todos os grupos clareados, a Vi celular foi reduzida em comparação à G1 (p< 0,05). Dentre os grupos clareados, G4 apresentou a menor quantidade de H2O2 difundido (p< 0,05), sendo que as células exibiram o menor EOx e a maior Vi (p<0,05). De maneira geral, G4 apresentou os maiores valores de EE (p<0,05). Conclusão: A estratégia de revestir o esmalte com SC+TiF4 antes da aplicação do gel clareador, reduz a CT e aumenta a EE do clareamento dental convencional de consultório.
