REGULACIÓN DE LOS MECANISMOS DE MOTILIDAD DE LAS CÉLULAS ADENOHIPOFISARIAS Y SU PAPEL EN LOS PROCESOS DE INVASIÓN DE LOS ADENOMAS HUMANOS.

- Abstract
Objetivo: Analizar la expresión de las integrinas alfa-2 (INTa2), beta-1 (INTß1) y beta-actina (ßACT) en las células tumorales adenohipofisarias, con el fin de averiguar la organización del citoesqueleto y conocer los posibles mecanismos de migración e invasión. Antecedentes: Los adenomas hipofisarios humanos (ADH) corresponden del 10-15% de los tumores intracraneales, del 25-55% son invasores y pueden ser agresivos. Las integrinas son proteínas transmembranales participan en la adhesión y señalización celular, su activación está mediada por actina; se han encontrado sobre expresadas en los ADH y su actividad está relacionada con la progresión de la enfermedad. Número de protocolo: 70-18 Métodos: Cortes histológicos de 4 µm de muestras de ADH e hipófisis normales incluidas en parafina fueron teñidos con H&E e inmunohistoquímica contra las hormonas adenohipofisarias (PRL, HC, FSH, LH, TSH, ACTH), INTa2, INTß1 y ßACT. Se analizó la positividad en un microscopio Nikon. Resultados: (Tabla 1) Se observaron 28 casos positivos (41.2%) a INTß1 (control sin positividad) y 47 (69.1%) a ßACT (control 2 casos positivos, 40%). Sin embargo, la INTa2 se encontró positiva en los controles (100%), que en los ADHs con 40 positivos (58.8%). Los I-ADH (contra los NI-ADH) mostraron más casos positivos a las tres proteínas, con 35 a INTa2 (57.4%), 26 a INTß1 (42.6%) y 40 a ßACT (65.6%). Los MADH (contra los GADH) también presentaron más casos positivos a las tres proteínas, con 36 a INTa2 (61%), 24 a INTß1 (40.7%) y 40 a ßACT (67.8%). Analizando la coexpresión de estas proteínas, se encontraron 8 positivos para INTß1 y ßACT, pero negativos para INTa2, de los cuales todos eran I-ADH y 7 eran MADH, 14 fueron positivos para las tres proteínas, de los cuales 11 eran MADH y 12 ADH invasores, y 7 fueron negativos para las tres proteínas, de los cuales todos eran I-ADH y 5 eran MADH. Conclusiones: Estudios recientes en modelos animales de adenomas hipofisarios han demostrado que la inhibición de la actividad de las integrinas y la actina puede tener efectos antitumorales. Nuestros resultados sugieren que la sobreexpresión de INTß1 y la ßACT podría estar asociada con mecanismos de movilidad, motilidad y capacidad invasiva de los ADH. En resumen, las tres proteínas parecen desempeñar un papel importante en la patogénesis de los adenomas hipofisarios, de ahí la importancia de conocer sus mecanismos de acción que permitirán desarrollar nuevas estrategias terapéuticas para su manejo médico.
