Comparación del efecto neuroprotector de dos formulaciones farmacéuticas de dapsona en un modelo de isquemia cerebral focal permanente en ratas.

- Abstract
Las enfermedades cerebrovasculares representan la tercera causa de morbi-mortalidad en el mundo. Se han desarrollado estrategias farmacológicas neuroprotectoras, sin resultados contundentes. Rios, et al, demostraron que la dapsona un fármaco con un mecanismo de acción capaz de inhibir los principales mecanismo de daño secundario desencadenados después de un evento isquémico cerebral (IC) mejora la función neurológica y preserva el tejido nervioso en ratas con IC, con base en esto, se realizó un estudio clínico en pacientes IC. Los resultados mostraron mayor recuperación en los pacientes tratados en comparación con el grupo placebo y la mayor recuperación se presento en los pacientes que recibieron dapsona mas tempranamente. Por lo que, es necesario desarrollar nuevas formulaciones farmacéuticas que sean utilizadas en situaciones de emergencia. Objetivos: desarrollar dos nuevas formulaciones farmacéuticas de dapsona y caracterizar los parámetros farmacocinéticos. Probar su efecto neuroprotector en un modelo de IC en rata utilizando escalas conductuales y cuantificando la cantidad de tejido dañado Material y métodos: se utilizaron 12 ratas con tratamiento intravenoso y 12 con formulación oral. Se muestreó sangre durante 24hs para determinar las concentraciones plasmáticas, biodisponibilidad y vida media. Segundo ensayo: se utilizaron 15 ratas con IC divididas en tres grupos: grupo-1, animales con IC+vehículo; grupo-2, IC+dapsona-v.o. y grupo-3, IC+dapsona-v.i. Los animales fueron evaluados a diferentes tiempos para determinar el daño neurológico utilizando escalas neurológicas. Resultados: no se observó toxicidad relacionada con la administración del fármaco, El estudio revela niveles máximo de 9.27 µg/ml/v.i. y 3.90 µg/ml/v.o. 4 hs después de su administración. Una biodisponibilidad del 87.6% y vidas medias similares. Se observó una mayor recuperación funcional en los animales tratados con ambas formulaciones vs control así como, menor cantidad de tejido dañado. Conclusiones: la dapsona demostró ser un fármaco neuroprotector efectivo en ambas formulaciones, esto se vio reflejado en un menor déficit neurológico y menor tejido dañado.
