- Abstract
Introducción: la neuropatía afecta a más del 50% de pacientes diabéticos. En 1990 se demostró la disminución de la concentración del factor de crecimiento neural (NGF) en pacientes con diabetes metlitus correlacionándose con la neuropatía diabética (ND), estos hallazgos fueron confirmados en modelos animales. El tratamiento con NGF en pacientes con ND tiene modestos resultados con altos costos. El ácido retinoico (AR) incrementa la expresión del NGF. Se demostró que el AR es capaz de prevenir la ND en ratones antes de estar establecida. El objetivo fue determinar si el AR es capaz de revertir la ND, cuando está se encuentra ya establecida. Se ultilizarón 90 ratones divididos en 3 grupos, A y B fueron tratados con estreptozotocina para inducir la DM. El grupo C como control. El grupo B fue tratado con 20mg/kg de AR por 30 días. A los 30 días y al finalizar se tomo la prueba de glucosa y sensibilidad térmico-nociceptivo de todos los grupos. Las concentraciones de NGF en suero y nervio ciático se determinaron por ELISA, se realizó un análisis ultraestructural del nervio para determinar la severidad delaND. Resultados: antes de el tratamiento con AR. El grupo A y B presentaron disminución significativa de la sensibilidad (figura 1). Estos cambios fueron revertidos con la administración de AR, 30 días después (figura 2). Los niveles de glucosa fueron mayores en el grupo A (279) B (264.5) en comparación con el C (86.8). El contenido de NGF en nervios y suero fue mayor en el grupo con DM, tratado con AR (2.4621 y 2.28 pg/µg) en comparación con el grupo con DM sin tratamiento (2.1778 y 1.9571 pg/µg) y (2.2800 y 2.4621 pg/µg) P= 0.0001 y P= 0.027. El daño ultaestructural fue menor en los animales diabéticos tratados con AR que en los no tratados. El AR aumenta las concentraciones de NGF y revlerte las alteraciones inducidas por ND.
