- Abstract
We demonstrate a stain-free classification of amyloid, collagen, and muscle fibers by high-sensitive detection of degree of linear polarization. A structure of an amyloid fiber is elongated with a diameter of several nm and a length of several m. On the other hand, fibers of collagens and muscles are strongly oriented. The orientation of these fibers gives us to yield their birefringence. For such birefringence properties, the degree of linear polarization deteriorates in cases of collagen and muscle fibers. We herein focus on the degree of linear polarization by each fiber, such as amyloid, collagen, and muscle. To detect their small changes of the degree of linear polarization by their fibers, we developed a high-sensitive detection of degree of linear polarization under a polarization microscope. We also obtained that the degree of linear polarization on amyloid fibers became 0.3% higher than that of muscle fibers. For this result, our high-sensitive detection of degree of linear polarization enables stain-free classification of amyloid, collagen, and muscle fibers. We expect that their results have a potential for detecting unstained amyloid.
