- Abstract
Using brewer's spent grains as raw materials, the medium for preparation of protein peptides by solid-state fermentation of Bacillus subtilis was studied. The effects of carbon source type and addition, solid to liquid ratio, potassium dihydrogen phosphate addition and water pH value on the yield of peptide were investigated by single factor experiments. The medium was optimized by Box-Behnken experiments and response surface analysis. The results showed that the optimal medium for fermentation of B. subtilis to produce brewer's spent grains protein peptide was pH 7.0, glucose addition 4.2%, wheat grains and water ratio 1.0∶2.1 (g∶ml), and potassium dihydrogen phosphate addition 0.6%. Under this condition, the yield of protein peptide fermented by brewer's spent grains could reach 12.40%.
