- Abstract
In order to identify pathogenic fungi causing cobweb disease in Oudemansiella raphanipes, a diseased fruiting body was subjected to tissue isolation and single spore isolation to obtain a pathogenic strain CG01. CG01 was then identified by its morphological characteristics and multigene phylogenetic analysis. For the morphological identification, CG01 was observed for its colony characteristics on PDA medium, and its microscopic characteristics of hyphae, conidiophores and conidia. For the phylogenetic analysis, sequences of ITS, RPB1, RPB2 and TEF were used. Based on the results of the above analyses and the Koch’s postulates, CG01 was confirmed to be a strain of Cladobotryum sp., with rather close consanguinity with C. paravirescens and C. asterophorum at the molecular level. The optimal temperature for the hyphal growth of CG01 was found to be 25 °C. This is the first report of pathogen responsible for cobweb disease in O. raphanipes.
