- Abstract
The experiment was to screen out a probiotic strain with fermented feed and bacteriostatic performance from the intestinal tract of cattle and sheep in Inner Mongolia, to analyze the biological characteristics of the strain. The results showed that one probiotic strain was screened and named as N9301. The biological characteristics were preliminarily determined as Streptococcus faecalis. Normal growth at pH value 4.2~9.2 could antagonize virulent Salmonella and virulent Escherichia coli. Through the analysis of antibacterial properties, there was determined that the ammonium sulfate saturation required for the crude bacteriocin of N9301 was 70%, and the antibacterial activity of the supernatant was the best after 16 h fermentation. The gel filtration peak solution was subjected to protein gel electrophoresis (SDS-PAGE) to prove that the molecular weight was less than 10 kDa. The crude extract of bacteriocin from Streptococcus faecalis N9301 had thermal stability (100 °C, 30 min), acid-base stability (pH value 2~1.3) and the antibacterial activity decreased after treatment with protease K, trypsin and pepsin, while α-amylase had no effect on antibacterial activity. The experiment indicates that Streptococcus faecalis N9301 has good bacteriostatic effect, and the bacteritin crude body fluid produced has strong acid-base stability and heat stability.
