- Abstract
A strain of dominant salt-tolerant bacteria NYTS1 was isolated from the activated sludge of a domestic wastewater treatment plant, which was identified as Pseudomonas by means of morphological observations, physiological and biochemical assays, and 16S r DNA sequence analysis. Then the parameters such as the amount of inoculum, glucose concentration, pH, salinity, dye concentration and temperature were selected as single factors to explore the impacts of different factors on NYTS1 strain growth and the removing of the dyestuff reducing Blue 4. Later, the main factors affecting the decolorization rate were screened by Plackett-Burman experiment, basing on which the conditions were optimized by the CCD experiment, and the decoloring validation experiments were performed according to the practical convenient correction conditions. Consequently, the experiments indicated: that the main factors affecting the decolorization rate were salinity, pH and temperature, and the descending order of influencing strength being pH>salinity>temperature. When the salinity was 2.78%, pH value was 7.33 and temperature was 35.72 ℃, the decoloring effect was the best, and the predicted rate being as high as 84.477 1%. The decoloration validation experiment showed the decoloring rate being equal to 83.24%, which was similar to the predicted value, indicating that it was feasible to optimize the decoloring conditions using the response surface methodology.
