- Abstract
Introdução: Membranas fibrilares (MFs) tem se destacado para a regeneração de tecidos craniofaciais, especialmente quando associados a moléculas bioativas capazes de estimular a neoformação de tecidos duros. Entretanto, o potencial odontogênico sobre células pulpares necessita ser melhor investigado, para que seja possível o desenvolvimento de biomateriais para utilização como capeadores pulpares diretos. Objetivo: Avaliar o potencial dentinogênico de membranas fibrilares de policaprolactona (PCL) incorporadas com hidróxido de cálcio (HC) sobre células pulpares humanas (HDPCs), visando desenvolver um biomaterial citocompatível e bioativo para aplicação sobre o tecido pulpar vital exposto. Método: Soluções de PCL (10% m/v) foram incorporadas com HC (PCL+HC; 0,5% m/v) ou não (PCL; controle) para confecção de MFs por eletrofiação. As MFs foram avaliadas quanto à morfologia e incorporação de HC por MEV/EDS, solubilidade por 6 meses e liberação de cálcio por 140 dias. HDPCs foram semeadas sobre as MFs e avaliadas quanto à viabilidade (alamarBlue e Live/Dead), adesão e espalhamento (F-actina), expressão gênica de marcadores da dentinogênese (RT-qPCR) e formação de matriz mineralizada (Alizarin Red) por até 21 dias. Os dados foram analisados com ANOVA e testes post-hoc (n=8; a=5%). Resultados: A incorporação de HC aumentou o diâmetro das fibras e a porcentagem de espaços interfibrilares. As MFs de PCL+HC demonstraram um padrão de degradação mais rápido que o controle, com liberação de cálcio significativa e constante. Aumento da viabilidade, adesão, espalhamento e expressão de ALPL, DSPP e DMP1 por HDPCs foi observada para PCL+HC, que aumentou o potencial de mineralização em até 9× mesmo sem suplementação odontogênica. Conclusão: Em conclusão, MFs de PCL+HC apresentaram perfil de degradação lento, liberação de cálcio constante, citocompatibilidade e estimularam a dentinogênese por HDPCs, sendo estratégias bioativas promissoras para aplicação sobre o tecido pulpar vital exposto.
