- Abstract
Introdução: A convergência dos conhecimentos no campo da medicina regenerativa e da engenharia de tecidos tem possibilitado a otimização do desenvolvimento de biomateriais destinados a servir como arcabouços (scaffolds) em uma ampla gama de aplicações, incluindo a reparação, regeneração e substituição de tecidos e órgãos. Um biomaterial amplamente empregado nesse contexto é o alginato de sódio, devido à capacidade de se transformar em gel de maneira eficiente e à habilidade de incorporar células e moléculas bioativas, as quais podem imprimir efeitos terapêuticos ao gel. O plasma rico em plaquetas (PRP), uma fração sanguínea que se caracteriza por conter concentrações elevadas de plaquetas e secretar fatores de crescimento e diferenciação celular, constitui-se em um excelente modelo de biomaterial, com grande potencial de aplicação na medicina regenerativa e translacional. Entretanto, o desenvolvimento de um gel que apresente propriedades essenciais para uso farmacológico, como biocompatibilidade e atoxicidade, é ideal para utilização terapêutica sem induzir efeitos colaterais no tecidoalvo. Objetivo: Nesse sentido, o presente estudo, teve por objetivo, avaliar os possíveis efeitos citotóxicos de um hidrogel resultante da associação de alginato e PRP. Método: Foram investigadas três diferentes concentrações de alginato 6%, 8% e 10% (v/v), em associação com Plasma Rico em Plaquetas (PRP) na proporção de 10:1 (v/v). Os hidrogéis resultantes foram fabricados utilizando uma bioimpressora (BioEnder©) e posteriormente submetidos a protocolos de esterilização por exposição à luz UV. Em seguida, as estruturas foram dispostas em placas de 12 poços. Cada uma das cavidades dessas placas foi então inoculada com células de hepatocarcinoma (HepG2/C3A). Após um período de incubação de 24 horas, foi realizado um teste de citotoxicidade empregando o ensaio de resazurina. Resultados: A leitura da placa de 12 poços realizada após 24h permitiu constatar que os poços estavam ausentes de contaminação com os hidrogéis de alginato a 10% e 6% associados ao PRP. Em adição, estes hidrogéis nas mesmas concentrações apresentaram viabilidade celular de 93% e 98%, respectivamente, demonstrando que os hidrogéis não induziram citotoxicidade às células. Conclusão: Os resultados obtidos permitem concluir que os hidrogéis de alginato a 8% e 10% são adequados para a bioimpressão 3D por extrusão, uma vez que apresentam viscosidade ideal para manter a conformação tridimensional dos scaffolds. Além disso, o teste de citotoxicidade mostrou que o hidrogel constituído de alginato e PRP não induziu citotoxicidade.
