Structures and ice-binding faces of the alanine-rich type I antifreeze proteins.

- Abstract
Antifreeze proteins (AFPs) protect cold-blooded organisms from the damage caused by freezing through their ability to inhibit ice growth. The type I AFP family, found in several fish species, contains proteins that have a high alanine content (>60% of the sequence) and structures that are almost all α-helical. We examine the structure of the type I AFP isoforms HPLC6 from winter flounder, shorthorn sculpin 3, and the winter flounder hyperactive type I AFP. The HPLC6 isoform structure consists of a single α-helix that is 37 residues long, whereas the shorthorn sculpin 3 isoform consists of two helical regions separated by a kink. The high-resolution structure of the hyperactive type I AFP has yet to be determined, but circular dichroism data and analytical ultracentrifugation suggest that the 195 residue protein is a side-by-side dimer of two α-helices. The alanine-rich ice-binding faces of HPLC6 and hyperactive type I AFP are discussed, and we propose that the ice-binding face of the shorthorn sculpin 3 AFP contains Ala14, Ala19, and Ala25. We also propose that the denaturation of hyperactive type I AFP at room temperature is explained by the stabilization of the dimerization interface through hydrogen bonds. Les protéines antigel (AFP, « antifreeze proteins ») protègent les organismes à sang froid des dommages causés par le gel grâce à leur capacité d’inhiber la croissance de la glace. La famille de protéines antigel AFP type I retrouvées chez plusieurs espèces de poissons, comprend des protéines à fort contenu en alanine (>60% de leur séquence) et leur structure est presque entièrement constituée d’hélices α. Nous examinons la structure de différentes isoformes d’AFP type I: la HPLC6 de la plie rouge, l’isoforme 3 du chaboisseau à épines courtes, et l’AFP type I hyperactive de la plie rouge. La structure de la HPLC6 consiste en une seule hélice α longue de 37 résidus, alors que l’isoforme 3 du chaboisseau à épines courtes consiste en deux régions hélocoïdales séparées par un coude. La structure à haute résolution de l’AFP type I hyperactive n’a pas encore été déterminée, mais les données obtenues par dichroïsme circulaire et par ultracentrifugation analytique suggèrent que la protéine de 195 résidus consiste en un dimère constitué de deux hélices α côte à côte. Les faces riches en alanines qui lient la glace de la HPLC6 et de l’AFP type I hyperactive font l’objet d’une discussion, et nous proposons que la face qui lie la glace de l’AFP 3 du chaboisseau à épines courtes contienne l’Ala 14, l’Ala19 et l’Ala25. Nous proposons aussi que la dénaturation de l’AFP type I hyperactive à température de la pièce s’explique par la stabilisation de l’interface de dimérisation via des ponts hydrogènes.
