- Abstract
Wild Thinopyrum grasses are important gene pools for forage and cereal crops. Knowledge of their chromosome organizations is pivotal for efficient utilization of this important gene pool in germplasm enhancement programs. Expressed sequence tags derived simple sequence repeat (EST-SSR) markers for Thinopyrum bessarabicum, T. elongatum, and T. junceum chromosomes were identified among amplicons produced from three series of wheat-Thinopyrum addition lines using 193 primer pairs designed from the Leymus EST unigenes. The homology of T. junceum chromosomes in 13 wheat addition lines was tentatively established to reveal that homologous groups 3, 4, 5, 6, and 7 were represented by HD3515, HD3505, AJDAj11, AJDAj1, and HD3508, whereas groups 1 and 2 were represented by AJADj7-AJDAj9 and AJDAj2-AJDAj4, respectively. AJDAj5 and AJDAj6 had complexly reconstituted T. junceum chromosomes that might have resulted from fusion or translocations of large chromosomal segments from two or more chromosomes, that is (1+5) and (2+5+1), respectively. The identified EST-SSR markers will be useful in comparative gene mapping, chromosome tracing, taxonomic studies, gene introgression, and cultivar identification. Les graminées sauvages du genre Thinopyrum constituent des réservoirs de gènes importants pour les cultures fourragères et céréalières. Une connaissance de leur organisation chromosomique s'avère essentielle pour une utilisation efficace de cette ressource pour l'amélioration du germoplasme. Des marqueurs EST-SSR pour les chromosomes du Thinopyrum bessarabicum, du T. elongatum et du T. junceum ont été identifiés au sein d'amplicons obtenus chez trois séries de lignées d'addition blé-Thinpyrum à l'aide de 193 paires d'amorces conçues sur la base d'unigènes EST provenant du genre Leymus. L'homologie des chromosomes du T. junceum chez 13 lignées d'addition a été établie provisoirement. Cette analyse a révélé que les groupes homologues 3, 4, 5, 6 et 7 étaient représentés chez les lignées HD3515, HD3505, AJDAj11, AJDAj1 et HD3508 tandis que les groupes 1 et 2 étaient représentés chez les lignées AJADj7 à AJDAj9 et AJDAj2 à AJDAj4 respectivement. Les lignées AJDAj5 et AJDAj6 contenaient des chromosomes du T. junceum reconstitués de manière complexe, potentiellement suite à des fusions ou translocations de grands segments chromosomiques provenant de deux chromosomes ou plus, par exemple (1+5) et (2+5+1) respectivement. Les marqueurs EST-SSR ainsi identifiés seront utiles en cartographie génétique comparée, pour le suivi des chromosomes, des études taxonomiques, l'introgression de gènes et l'identification de cultivars.
