- Abstract
To study the effects of Fructus Ligustri Lucidi on the proliferation and differentiation of cultured osteoblasts in vitro, the proliferation and the activity of alkaline phosphatase (ALP) in osteoblastic UMR-106 cells was investigated in the presence of water extract of Fructus Ligustri Lucidi (LWE) with Am-blue method and p-nitrophenyl phosphate disodium salt (PNPP) method respectively. The results showed that LWE promoted the proliferation and differentiation of UMR-106 for 48 h at 100 µg/mL. To study the mechanism of LWE on differentiation, pGL3-5 x ERE-Luc reporter mid was transfected into breast cancer cell MCF-7 ,and the effect of LWE on the expression of luciferase was investigated. The results indicated that LWE promoted the estrogen receptor responsive element (ERE) driven expression of luciferase. The promoting effect of LWE on the differentiation of UMR-106 cells could be suppressed by ICI182780, an estrogen receptor antagonist. These results suggested that the mechanism of differentiation promoting effect by LWE may be related to estrogen receptor signaling pathway.
