- Abstract
Helminthiasis is a serious problem in tropical and subtropical region including India, in which soil transmitted helminthiasis (STH) is more frequent. Schleichera oleosa, known as 'Kusum' or 'Lac Tree', is well known deciduous tree used traditionally as anthelmintic. The present investigation includes the isolation, characterization, in-vitro anthelmintic effect of isolated compound (quercetin) on Ascardia galli. Successive extraction of S. oleosa leaf with different solvents was evaluated qualitatively and quantitatively. Ethanolic extract (SOEE) was subjected for TLC analysis and HPTLC fingerprinting.Isolation was carried out by column chromatography. Characterization of the isolated compound was done by IR, ¹H NMR and Mass spectroscopy. Isolated compound (SOEE/C-1) was evaluated against A. galli (collected from freshly slaughtered domestic Galus gallus) to validate the anthelmintic potency. Identification of flavonoids, tannins and phenolics were quantified in which SOEE was more abundant. TLC analysis with different solvents (system I and II) helped to analyze the Rf value of visualized spots. Rf value matched with standard quercetin (Rf value 0.9' and 0.65). In HPTLC fingerprinting quercetin was affirmed (Rf value 0.49, Band no 4) while comparing with standard quercetin. Eluents (fraction A,B,C and D) collected from column chromatography; the fraction C (ethyl acetate: ethanol-80:20 ratio) was subjected for TLC (single spot, Rf value 0.91, solvent system I). The data obtained from spectroscopic analysis of the isolated compound (SOEE/C-1) was evaluated, and quercetin was confirmed. The traditional use of the plant S. oleosa as anthelmintic was hence proved here, along with the isolation of quercetin from the ethanolic extract was done and characterized. The exact mode of action of quercetin on A. galli will be the future scope.
