EFEITOS DA SUPLEMENTAÇÃO COM EXTRATO HIDROALCÓOLICO DE Euterpe oleracea MART. (AÇAÍ) SOBRE ALTERAÇÕES CARDIOVASCULARES NA MENOPAUSA EXPERIMENTAL.

- Abstract
Introdução: As alterações hormonais relacionadas à menopausa, principalmente a queda dos níveis de estrogênio, levam ao risco do desenvolvimento de doenças cardiovasculares. A terapia de reposição hormonal é frequentemente usada para aliviar as consequências clínicas da menopausa. No entanto, parece estar relacionada ao aparecimento de câncer de mama e tromboembolismo. Estudos do nosso grupo de pesquisa têm demonstrado que o extrato hidroalcoólico de Euterpe oleracea MART. (Açaí Seed Extract - ASE), rico em compostos polifenólicos, apresenta efeitos cardiovasculares benéficos em modelos experimentais de hipertensão. Objetivo: Avaliar os efeitos benéficos do ASE sobre as alterações cardiovasculares em modelo experimental de menopausa. Método: A menopausa experimental foi induzida por ovariectomia bilateral (OVX) em ratas Sprague-Dawley (90 dias de idade) sob anestesia com isoflurano (4% para hipnose + 1,0 L/min de O2; 2% para manutenção + 0,8 L/min de O2). Ratas Sham foram usadas como controles. Um mês após a cirurgia, o tratamento com ASE (200 mg/Kg/dia por gavagem intragástrica) foi iniciado e durou 8 semanas. Quatro grupos experimentais foram utilizados: SHAM, SHAM+ASE, OVX e OVX+ASE. O peso corporal (n=13) foi avaliado semanalmente durante o período experimental. A pressão arterial sistólica (PAS) (n=6) foi aferida semanalmente por pletismografia de cauda. Ao final do tratamento, as ratas foram eutanasiadas por decapitação, o leito arterial mesentérico (LAM) foi isolado para avaliar sua responsividade a agentes vasoativos (n=6-7), e a aorta torácica foi coletada para análise morfométrica, a fim de avaliar a espessura da túnica média (n=5-6) e a fibrose a partir da coloração Picrossirius Red (n=5-7). Todos os protocolos experimentais foram aprovados (CEUA IBRAG/UERJ: n° 034/2022). Resultados: Ao final do tratamento, o grupo OVX apresentou aumento do peso corporal quando comparado ao grupo SHAM, enquanto o tratamento com ASE diminuiu esse parâmetro nos grupos SHAM+ASE e OVX+ASE em relação aos respectivos controles A PAS foi maior no grupo OVX quando comparada ao grupo SHAM+ASE. Além disso, o grupo OVX+ASE apresentou diminuição da PAS quando comparado ao grupo OVX A resposta vasoconstritora induzida por noradrenalina no LAM não diferiu significativamente entre os grupos. No entanto, a resposta vasodilatadora induzida pela acetilcolina foi menor no grupo OVX do que no grupo SHAM, e a suplementação com ASE não afetou esse parâmetro. Além disso, a espessura da túnica média, assim como a fibrose da aorta torácica foi maior no grupo OVX quando comparada ao grupo SHAM, e a suplementação com ASE diminuiu esses parâmetros. Conclusão: Os resultados sugerem que a suplementação com ASE previne a elevação da pressão arterial sistólica, a fibrose vascular e o remodelamento vascular associados à menopausa experimental. No entanto, ainda são necessários estudos para aprofundar o conhecimento sobre os efeitos benéficos do extrato, visando prevenir ou atenuar as consequências cardiovasculares da menopausa humana.
