Keywords: GBM; DIPG; diffuse intrinsic pontine glioma; novel therapy; blood-brain barrier; drug delivery; targeting High grade gliomas including glioblastoma (GBM) in adults and diffuse intrinsic pontine glioma (DIPG) in children are fatal brain tumors with <5% of patients surviving 5 years after initial diagnosis and treatment. While promising results have generated enthusiasm for CAR-T cell therapy of brain tumors, expanded search for CAR targets, improved trafficking and optimization of dose, frequency and schedule of administration, will be key to advancement of CAR-T cell therapy.
