- Abstract
Considering the importance of milk in the diet of Brazilian people and its restriction to patients with phenylketonuria, this study aimed to reduce the phenylalanine (Phe) content of this food. Thus, several enzymatic hydrolysates were prepared, using a protease from Bacillus subtilis and the effect of some parameters were tested, such as E:S ratio, time and temperature of the reaction. Phe removal was performed using activated carbon (AC) as adsorbent support, and the protein:CA ratios of 1:22, 1:44 and 1:88 were evaluated. The efficiency of Phe removal was estimated by second derivative spectrophotometry, determining the Phe content in milk and in its hydrolysates, after treatment with AC. The highest Phe removal (75.9%) was achieved using a reaction time of 5h, a temperature of 50°C, an E:S ratio of 4:100 and a protein:AC ratio of 1:22. This assay resulted in a milk with a final Phe content of 36.9 mg /100 ml, which would allow its use by patients with phenylketonuria.
