- Abstract
Introdução: A catálise com óxidos metálicos tem despontado como uma excelente alternativa para aumento da eficácia estética e redução dos danos tóxicos causados pelo clareamento dental de consultório, entretanto, ainda não se sabe como a catálise com esses compostos podem atuar combinados com géis clareadores a base de peróxido de carbamida. Objetivos: Avaliar a influência da adição de óxido de manganês (MnO) num gel clareador contendo 22% de peróxido de carbamida (PC), sobre a citotoxicidade trans-amelodentinária (CT), eficácia estética (EE) e cinética de degradação (CD). Material e Métodos: Discos padronizados de esmalte/dentina, adaptados com câmaras pulpares artificiais, foram distribuídos de acordo com os tratamentos, estabelecendo os seguintes grupos (n=8): G1 - Sem tratamento (controle negativo); G2 - 22%PC (controle positivo - Clareador Opalescence 22%, Ultradent); G3 - 22%PC + 2mg/mL de MnO; G4 - 22%PC + 6mg/mL de MnO; G5 - 22%PC + 10mg/mL de MnO. A EE (ΔWI) dos tratamentos propostos foi determinada após 5, 10 e 15 dias (ANOVA/Sidak). Para avaliação da CT, os extratos (meio de cultura + componentes dos géis difundidos pelos discos) foram aplicados sobre células MDPC-23, as quais foram analisadas quanto a viabilidade (Vi) e estresse oxidativo (EOx). A difusão trans-amelodentinária de H2O2 (DTH2O2) foi determinada. A CD foi avaliada através da quantidade de radical hidroxila (OH-) formado (ANOVA/Tukey; p>0,05) Resultados: G5 apresentou os maiores valores de EE em todos os períodos, quando comparado a G2 (p0,05).) A menor DTH2O2 aconteceu em G5, onde as células exibiam a maior Vi e o menor EOx (p<0,05). Comparado a G2, aumento na produção de OH-ocorreu nos grupos onde MnO foi usado (p<0,05), sendo os maiores valores observados em G5 (p<0,05). Conclusão: A incorporação de 10 mg/mL de MnO num gel contendo 22% de PC aumenta a EE e a CD, minimizando a CT do produto.
